Mechanism and a new method for monitoring lysophagy
Autophagy is a degradation system within a cell. Cytoplasmic materials are sequestered by autophagosomes and delivered to lysosomes for degradation. We have found that the lysosome is selectively sequestered by autophagy when its membrane is damaged; this process has been called “lysophagy”, but the mechanism behind it is still unclear. Recently, besides autophagy, it has been reported that pathways mediated by the ESCRT complex and the transcription factor TFEB act to repair damaged lysosomes. Because it is difficult to evaluate these pathways and the lysophagy separately, we have established a new evaluation system that specifically monitors lysophagy. Moreover, we have identified candidate factors related to lysophagy with our own screening system, and we are investigating their involvement in lysophagy.
