Visualization of protocadherin cell-cell interaction

In mice, 58 clustered protocadherins are expressed in neurons, but approximately 10 isoforms are stochastically expressed in each cell. The combination of expressed protocadhereins is thought to give neurons identity. Protocadherins interact with each other in trans, but their functions are unknown. We created a probe that visualizes the interaction using FRET. When we observed the FRET probes in neurons, we found that the interaction occurs at non-synaptic sites.
