Regulation of Sox2 gene in early sensory development

Transcriptional factor Sox2 plays a central regulatory role in various stem cells, neural and sensory development. The Sox2 is regulated by ~30 distinct enhancers in the several 100 kb region surrounding the gene with distinct temporal and special specificities. Among these enhancers, NOP1 enhancer is active in the inner ear in its early developmental stages. We analyzed this enhancer to elucidate the mechanisms underlying the inner ear development. The NOP1 enhancer is activated by the synergistic action of Sox2 and Sall4, but the activation mechanisms do not generate its specificity. It is repression mechanisms partially mediated by Zeb1/2 that confine the enhancer activity to the inner ear. It is possible that these counteracting mechanisms operate in the tissue specification of the inner ear.
