Yeast prions provide a well characterized model system for studying the deposition of amyloid aggregates in cells. Aggregates of [PSI+] and [RNQ+] prions have been studied by electron tomography of both freeze-substituted and vitrified sections of yeast cells. The prion fibrils form partially ordered arrays which are not bounded by any membranes, but which exclude ribosomes and other visible cellular complexes. The fibril organization depends on the presence of the Hsp70 chaperone system, and sub-tomogram averages reveal a network of cross bridging structures between the fibrils.
